RESUMEN

Temporary immersion systems (TIS) are technological tools that support plant micropropagation. Given their high efficiency in the in vitro propagation of shoots, a current goal is to update the protocols addressing micropropagation in semisolid culture systems to protocols involving TIS. To this end, different parameters have been evaluated, including TIS types and designs, immersion times, immersion frequencies, and volume of medium per explant, among other characteristics. This has resulted in the improved production of propagules of plants of economic interest and the production of physiologically upgraded plants with high percent survival during acclimatization. TIS are specialized culture flasks that provide countless advantages during the commercial micropropagation of plants.

RESUMEN

The cultivation of vanilla (Vanilla planifolia) is of economic interest because vanillin is extracted from the fruits of this species. Vanillin is a natural flavoring highly valued in the food market. However, there is a short supply of propagules available for establishing commercial plantations and good-quality plants with phytosanitary certification. Plant tissue culture represents a viable option to supply large amounts of healthy plants to vanilla producers. In addition, the use of temporary immersion systems will allow commercial scale-up and the establishment of biofactories dedicated to in vitro vanilla propagation. This chapter describes a large-scale micropropagation protocol for vanilla using temporary immersion bioreactors (TIB).

RESUMEN

Marine species, such as corals and kelp, are responding to climate change by altering their distributions. Microbial biofilms underpin key processes that affect the establishment, maintenance, and function of these dominant habitat-formers. Climate-mediated changes to microbial biofilms can therefore strongly influence species' range shifts. Here, we review emerging research on the interactions between benthic biofilms and habitat-formers and identify two key areas of interaction where climate change can impact this dynamic: (i) via direct effects on biofilm composition, and (ii) via impacts on the complex feedback loops which exist between the biofilm microbes and habitat-forming organisms. We propose that these key interactions will be fundamental in driving the speed and extent of tropicalisation of coastal ecosystems under climate change.

Abstract Endozoochory by waterbirds is particularly relevant to the dispersal of non-flying aquatic invertebrates. This ecological function exercised by birds has been demonstrated in different biogeographical regions, but there are no studies for the neotropical region. In this work, we identified propagules of invertebrates in faeces of 14 syntopic South American waterbird species representing six families, and hatched additional invertebrates from cultured faeces. We tested whether propagule abundance, species richness and composition varied among bird species, and between the cold and warm seasons. We found 164 invertebrate propagules in faecal samples from seven different waterbirds species, including eggs of the Temnocephalida and Notonectidae, statoblasts of bryozoans (Plumatella sp.) and ephippia of Cladocera. Ciliates (including Paramecium sp. and Litostomatea), nematodes and rotifers (Adineta sp. and Nottomatidae) hatched from cultured samples. Potential for endozoochory was confirmed for 12 of 14 waterbird species. Our statistical models suggest that richness and abundance of propagules are associated with bird species and not affected by seasonality. Dispersal by endozoochory is potentially important to a broad variety of invertebrates, being promoted by waterbirds with different ecological and morphological traits, which are likely to drive the dispersal of invertebrates in neotropical wetlands.

Premise: Variation in seed traits is common within and among populations of plant species and often has ecological and evolutionary implications. However, due to the time-consuming nature of manual seed measurements and the level of variability in imaging techniques, quantifying and interpreting the extent of seed variation can be challenging. Methods: We developed a standardized high-throughput technique to measure seed number, as well as individual seed area and color, using a derived empirical scale to constrain area in Arabidopsis thaliana, Brassica rapa, and Mimulus guttatus. We develop a specific rational model using seed area measured at various spatial scales relative to the pixel count, observing the asymptotic value of the seed area as the modeled number of pixels approaches infinity. Results: We found that our model has high reliability in estimating seed traits and efficiently processes large numbers of images, facilitating the quantification of seed traits in studies with large sample sizes. Discussion: This technique facilitates consistency between imaging sessions and standardizes the measurement of seed traits. These novel advances allow researchers to directly and reliably measure seed traits, which will enable tests of the ecological and evolutionary causes of their variation.

The coastal saltmarshes in China have been seriously degraded and artificial restoration strategies have become one of the primary measures to protect the coastal habitats. In this study, we investigated efficient restoration techniques for native saltmarsh plants in vacant habitats within the project area of the Ecological Control of Spartina alterniflora and Improvement of Birds Habitats in the Shanghai Chongming Dongtan Bird National Nature Reserve in the Yangtze Estuary. Through field and pilot experiments, we analyzed the effects of propagule types and transplanting methods on the restoration efficiency of Scirpus mariqueter, a dominant saltmarsh species in the Yangtze Estuary. We further evaluated the cost and efficiency of various restoration strategies. Our results indicated that: 1) The corm seedlings of S. mariqueter had higher flood tolerance, and were suitable for use in the habitats with higher environmental heterogeneity. 2) Across the four treatments for S. mariqueter restoration, the most economically efficient treatment was to transplant low-density corm seedlings without sediment, with an investment cost of approximately ï¿¥10100 per hectare. The costs were ï¿¥41100, ï¿¥30000, and ï¿¥120100 per hectare for high-density without sediment, low-density with sediment, and high-density with sediment restoration, respectively. 3) After nearly 5-yr efforts, the S. mariqueter community of Plot C3 in project area had achieved a coverage of over 60%, marking a relatively successful large-scale field restoration. This study could provide the foundation and support for large-scale coastal saltmarsh restoration projects.

Submerged macrophytes play a key role in the restoration of shallow eutrophic lakes. However, in some subtropical lakes, benthivorous fishes dominate the fish assemblages and influence the growth of submerged plants. A comprehensive understanding of the direct and indirect effects of benthivorous fishes on submerged plants is important. We conducted mesocosm experiments to examine the effects of three densities of benthivorous fish, Misgurnus anguillicaudatus, on the water properties, the growth, asexual reproduction, and the germination of turions of Potamogeton crispus L. Our results showed that fish disturbance increased TN, TP, PO4-P, NH4-N, and NO3-N of the water, raising the extinction coefficient K, Chl a, and the periphyton biomass. Benthivorous fish disturbance reduced the total biomass, root length, relative growth rate (RGR), and branching number while increasing the plant height of P. crispus. The P stoichiometric homeostasis coefficient (H P) (except turions) and H N was lower in plant tissues due to fish disturbance. Benthivorous fish disturbances promoted turions formation (e.g., increased turions total numbers and biomass) of P. crispus. Moreover, P. crispus exhibited transgenerational plasticity for benthivorous fish affecting turion emergence. The maximum final germination rate occurred only when fish density in the mother plant grow experiment matched that in the turion germination experiment. Turions generated by P. crispus disturbed by low-density fish exhibited increased germination rates. Our findings suggest that controlling benthivorous fish reduces its indirect and direct effects on submerged vegetation, facilitating the successful restoration of these plants.

Environmental biomonitoring is a prerequisite for efficient evaluation and remediation of ecosystem degradation due to anthropogenic pressure or climate change. Estuaries are key habitats subject to multiple anthropogenic and natural stressors. Due to these multiple stressors, the detection of anthropogenic pressure is challenging. The fact that abundant natural stressors often lead to negative quality assessments has been coined the "estuarine quality paradox". To solve this issue, the application of molecular approaches with successful bioindicators like foraminifera is promising. However, sampling protocols, molecular procedures and data analyses need to be validated before such tools can be routinely applied. We conducted an environmental DNA survey of estuarine mudflats along the French Atlantic coast, using a metabarcoding approach targeting foraminifera. Our results demonstrate that estuarine environments have only a few active OTUs dominating the community composition and a large stock of dormant or propagule stages. This last genetic diversity components constitute an important reservoir, with different species which can potentially develop in response to the temporal variability of the multiple stressors. In fact, different OTUs were dominant in the studied estuaries. Our statistical model shows that the physical and chemical characteristics of the sediment and the climatic conditions explain only 43 % of the community composition variance. This suggests that other, less easily quantifiable factors, such as the history and use of the estuaries or the ecological drift could play an important role as well. Environmental DNA biomonitoring opens new perspectives to better characterize the genetic diversity in estuaries.

Trichoderma spp. are a genus of well-known fungi that promote healthy growth and modulate different functions in plants, as well as protect against various plant pathogens. The application of Trichoderma and its propagules as a biological control method can therefore help to reduce the use of chemical pesticides and fertilizers in agriculture. This review critically discusses and analyzes groundbreaking innovations over the past few decades of biotechnological approaches to prepare active formulations containing Trichoderma. The use of various carrier substances is covered, emphasizing their effects on enhancing the shelf life, viability, and efficacy of the final product formulation. Furthermore, the use of processing techniques such as freeze drying, fluidized bed drying, and spray drying are highlighted, enabling the development of stable, light-weight formulations. Finally, promising microencapsulation techniques for maximizing the performance of Trichoderma spp. during application processes are discussed, leading to the next-generation of multi-functional biological control formulations. KEY POINTS: • The development of carrier substances to encapsulate Trichoderma propagules is highlighted. • Advances in biotechnological processes to prepare Trichoderma-containing formulations are critically discussed. • Current challenges and future outlook of Trichoderma-based formulations in the context of biological control are presented.

Fungi can colonize organic matter present in subterranean sites and have a significant role as dwellers in different microniches of cave habitats. In order to analyze the content of airborne fungal propagules in different parts of "Stopica Cave," a touristic site in Serbia, air sampling was carried out in three seasons during 2020, prior to and during the onset of COVID-19 pandemic. Culturable mycobiota was identified using both microscopic techniques and ITS region/BenA gene barcoding, while multivariate analyses were employed to establish the link between fungal taxa and different environmental factors. The maximal measured fungal propagule concentrations were recorded during spring sampling which were based on fungal propagule concentration categories; the cave environment matches the category V. A total of 29 fungal isolates were identified, while Aspergillus, Cladosporium, Fusarium, Lecanicillium, Mucor, and Penicillium were the most diverse genera. According to the trophic mode, most of the isolated fungal species were pathotrophs (75.86%), but when regarding ecological guilds, the most dominant were undefined saprobes and animal pathogens (41.38% for each). Show caves are especially vulnerable to human impacts, and the fungal propagules' concentration within the caves could be good indices for the level of ecological disturbance.

Orchids produce large numbers of dust-like seeds that rely heavily on orchid mycorrhizal fungi (OMFs) for germination. Using OMFs to facilitate orchid proliferation is considered an effective method for orchid conservation but still presents challenges in practice. In this study, orchid seed-fungus complexes, in which orchid seeds and fungal mycelia were embedded together to form granules, were developed as platforms to facilitate seed germination and seedling production. Overall, seedlings were produced by seed-fungus complexes for five orchid species with large variations in the percentages of seedlings produced among species/treatments. For the different fungal treatments in Dendrobium officinale, Sebacinales LQ performed much better than the other fungal strains. At 90 days after sowing, 75.8±2.6% seedlings were produced in the LQ treatment, which was significantly higher than in the Tulasnella sp. JM (22.0±3.0%) and Tulasnella sp. TPYD-2 (5.3±1.0%) treatments, as well as in the LQ and TPYD-2 cocultured treatment (40.4±3.2%), while no seedlings were formed in the Tulasnella sp. SSCDO-5 or control treatments. For the other four orchid species, only one compatible fungus for each species was used, and the percentages of seedlings in epiphytic Dendrobium devonianum (67.2±2.9%) and D. nobile (38.9±2.8%) were much higher than those in terrestrial Paphiopedilum spicerianum (2.9±1.1%) and Arundina graminifolia (6.7±2.1%) at 90 days after sowing. Adding 1% polymer water-absorbent resin to the seed-fungus complexes of D. officinale seeds with fungal strain Sebacinales LQ significantly increased seedling formation, while other additional substances showed negative effects on seedling formation. For the storage of seed-fungus complexes, it is recommended to store the seed-fungus complexes in valve bags at room temperature for a short time and at a low temperature of 4°C for no more than 30 days. As a platform for symbiotic seed germination, the seed-fungus complex can facilitate seed germination, produce seedlings and support subsequent seedling growth, and its seedling productivity depends on seed germination characteristics, seed viability, and the efficiency of fungi. Seed-fungus complexes have great potential to be used as propagules in orchid conservation.

The ecological importance of phytoplankton-benthic-propagules (PBP) from coastal sediments, except tropical monsoon-influenced rivers and estuaries, is well documented. PBP in such systems is often transported from upstream (near freshwater) to downstream (estuary) through freshwater discharges during monsoon and thereby experiences higher salinities (>30 PSU) and nutrients with varying light conditions due to reducing discharges during the monsoon-break/withdrawal-phase. However, the PBP responses (germination and subsequent growth) to such changes are unknown and are examined here at ~35 PSU salinity. For the study, macronutrients (nitrate, phosphate, silicate, and nitrate+phosphate+silicate) and light intensities were considered to assess the response of PBP representing estuarine, brackish, and near freshwater locations of monsoon-influenced Mandovi and Zuari rivers (Goa, India). Diatoms dominated the viable PBP community, but the maximum growth and sustained photosynthetic activity were observed when all macronutrients were supplied than individually. Here, variable fluorescence technique utility in PBP resurrection (detection of viability and responses) was also explored. The PBP growth was similar for macronutrients but increased with light intensity indicating a longer growth response during monsoon. For PBP (germination and photosynthetic activity), light intensities drive the rate of improvement/development, whereas the nutrients are essential for maintaining vegetative growth upon germination in the region. The PBP dominance of common planktonic species (Skeletonema and Thalassiosira) along the river also signifies the role of seawater intrusion (up to upstream locations) in distribution. Skeletonema and Thalassiosira, which contribute significantly to the total community, are light-sensitive with a similar response and cause single species blooms during monsoon and non-monsoon, respectively, depending on the species' tolerance to salinity.

In vitro propagation, also known as micropropagation, has become the most widely employed method for blackberry propagation, as it overcomes the limitations of the traditional asexual propagation methods (mainly layering and cutting). In this context, synthetic seed technology represents a strategy to enhance the productivity of in vitro propagation and facilitates the exchange of plant materials between laboratories, contributing to germplasm conservation. This study aimed to identify the most suitable vegetative propagule for the encapsulation of blackberry. To this end, uninodal microcuttings (nodes) and the base of clumps were used to produce synthetic seeds for the cultivars Thornfree and Chester. Forty-five days after sowing, viability (percentage of green propagules without browning or necrosis), regeneration (percentage of propagules that sprouted and rooted simultaneously), number of shoots produced, shoot length, number of roots produced, root length, and the fresh and dry weights of the plantlets were measured. The results demonstrated that both considered propagules allowed us to obtain satisfactory regeneration rates. However, plantlets originating from the encapsulated clump's base had more shoots and roots, resulting in greater fresh and dry weights than the plantlets derived from encapsulated nodes. Therefore, for achieving more robust plantlets and enhancing overall procedural efficiency, we recommend using the base of clumps as a propagule for blackberry encapsulation.

Aedes aegypti (Linn.) incidence has increased in recent years, causing human viral diseases such as dengue, which are often fatal. Beauveria bassiana (Bals., Vuillemin) efficacy for Ae. aegypti biological control has been evidenced but it relies on host susceptibility and strain virulence. We hypothesized that B. bassiana conidia microgranular formulations (MGF) with the additives acetone, lactic acid, and sugar increase Ae. aegypti adult exposure, thus improving their biocontrol effectiveness. Beauveria bassiana strain four (BBPTG4) conidia stability was assessed after 0 d, 5 d, and 30 d storage at 25 °C ± 2 °C with additives or in MGF after 91 d of storage at 25 °C ± 2 °C or 4 °C ± 1 °C, whereas mortality was evaluated after adult exposure to MGF + conidia, using home-made traps. Additives did not show toxicity to conidia. In addition, we observed that sugar in MGF increased Ae. aegypti adults' attraction and their viability resulted in a 3-fold reduction after 5 d and 1- to 4-fold decrease after 30 d of storage, and formulations were less attractive (p < 0.05). Conidia stability was higher on MGF regardless of the storage temperature, losing up to 2.5-fold viability after 91 d. In conclusion, BBPTG4 infected and killed Ae. aegypti, whereas MGF attracting adults resulted in 42.2% mortality, increasing fungus auto dissemination potential among infected surviving adults. It is necessary to further evaluate MGF against Ae. aegypti in the field.

The complex morphological structure of Aspergillus niger influences its production of proteins, metabolites, etc., making the genetic manipulation and clonal purification of this species increasingly difficult, especially in aconidial Aspergillus niger. In this study, we found that N-acetyl-D-glucosamine (GlcNAc) could induce the formation of spore-like propagules in the aconidial Aspergillus niger SH2 strain. The spore-like propagules possessed life activities such as drug resistance, genetic transformation, and germination. Transcriptomic analysis indicated that the spore-like propagules were resting conidia entering dormancy and becoming more tolerant to environmental stresses. The Dac1 gene and the metabolic pathway of GlcNAc converted to glycolysis are related to the formation of the spore-like propagules, as evidenced by the CRISPRi system, qPCR, and semi-quantitative RT-PCR. Moreover, a method based on the CRISPR-Cas9 tool to rapidly recycle screening tags and recover genes was suitable for Aspergillus niger SH2. To sum up, this suggests that the spore-like propagules are resting conidia and the mechanism of their formation is the metabolic pathway of GlcNAc converted to glycolysis, particularly the Dac1 gene. This study can improve our understanding of the critical factors involved in mechanisms of phenotypic change and provides a good model for researching phenotypic change in filamentous fungi.

The process of plastids developing into chloroplasts is critical for plants to survive. However, this process in woody plants is less understood. Kandelia obovata Sheue, Liu & Yong is a viviparous mangrove species; the seeds germinate on the maternal tree, and the hypocotyls continue to develop into mature propagules. We identified rare albino propagules through field observation among normal green and brown ones. Toward unveiling the propagule plastid development mechanism, albino propagule leaves only have etioplasts, low photosynthesis rates, and drastically reduced chlorophyll a/b and carotenoid contents, but with increased superoxide dismutase activities. To identify candidate genes controlling propagule plastid development, a genome-wide association study (GWAS) was performed between the albino and green propagules. Twenty-five significant single nucleotide polymorphisms (SNPs) were associated with albino propagule plastid development, the most significant SNPs being located on chromosomes 1 and 5. Significant differentially expressed genes were identified in porphyrin and chlorophyll metabolisms, carotenoid and flavonoid biosynthesis by combining transcriptome and GWAS data. In particular, KoDELLAs, encoding a transcription factor and KoCHS, encoding chalcone synthase, may be essential to regulate the albino propagules plastid development through weakened chlorophyll and flavonoid biosynthesis pathways while promoting chlorophyll degradation. Our results provide insights into genetic mechanisms regulating propagule plastid development in woody plants.

Nymphaea, commonly known as water lily, is the largest and most widely distributed genus in the order Nymphaeales. The importance of Nymphaea in wetland ecosystems and their increased vulnerability make them a great choice for conservation and management. In this work, we studied genetic diversity in a collection of 90 N. micrantha and 92 N. nouchali individuals from six different states of India, i.e., Assam, Manipur, Meghalaya, Maharashtra, Goa, and Kerala, using simple sequence repeat (SSR) markers developed by low throughput Illumina sequencing (10X coverage of genome) of N. micrantha. Nymphaea nouchali is native to India, whereas N. micrantha is suggested to be introduced to the country for its aesthetic and cultural values. The study revealed extensive polymorphism in N. nouchali, while in N. micrantha, no apparent genetic divergence was detected prompting us to investigate the reason(s) by studying the reproductive biology of the two species. The study revealed that N. micrantha predominantly reproduces asexually which has impacted the genetic diversity of the species to a great extent. This observation is of immense importance for a successful re-establishment of Nymphaea species during restoration programs of wetlands. The information generated on reproductive behaviors and their association with genotypic richness can help in strategizing genetic resource conservation, especially for species with limited distribution. The study has also generated 22,268 non-redundant microsatellite loci, out of which, 143 microsatellites were tested for polymorphism and polymorphic markers were tested for transferability in five other Nymphaea species, providing genomic resources for further studies on this important genus.

Interventions with commercial inoculants have the potential to reduce the environmental footprint of agriculture, but their indiscriminate deployment has raised questions on the unintended consequences of microbial invasion. In the absence of explicit empirical reports on arbuscular mycorrhizal fungi (AMF) invasion, we examine the present framework used to define AMF invasion and offer perspectives on the steps needed to avoid the negative impacts of AMF invasion. Although commercial AMF isolates are potential invaders, invasions do not always constitute negative impacts on native community diversity and functions. Instead, the fates of the invading and resident communities are determined by ecological processes such as selection, drift, dispersal, and speciation. Nevertheless, we recommend strategies that reduce overdependence on introduced inoculants, such as adoption management practices that promote the diversity and richness of indigenous AMF communities, and the development of native propagules as a supplement to commercial AMF in applicable areas. Policies and regulations that monitor inoculant value chains from production to application must be put in place to check inoculant quality and composition, as well as the transport of inoculants between geographically distant regions.

Riparian forests suffered from forest fragmentation, which in turn promote the change in the pattern of seed dispersion and modification of soil seed bank.  The aim of this study is to indicate the main species that make up the seed bank of a stretch of riparian forest of the Mogi Guaçu River continued to a fragment of semidecidual forest, located in the Guarnição da Aeronáutica de Pirassununga. It was delimited ten plots of 10 x 10 m. Four soil samples was collected by plot with a 25 x 25 cm template. Samples were packed in greenhouse for seed germination and monitored every 15 days for six months. Altogether, 494 individuals from 19 families were sampled, distributed in 30 genera and 40 morphospecies. Diversity and distribution of species in the fragment is homogeneous and can be used as source of propagules for forest restoration.


Florestas ripárias sofrem com a fragmentação florestal, que promove mudanças no padrão de dispersão de sementes e modificação do banco de sementes. O objetivo do presente estudo é indicar as principais espécies que compõem o banco de semntes de um trecho de floresta ripária do Rio Mogi Guaçu River contínuo a um fragmento de floresta estacional, localizada na Guarnição da Aeronáutica de Pirassununga. Foram delimitadas dez parcelas de 10 x 10 m. Quatro amostras de solo foram coletadas através de um molde de 25 x 25 cm. Amostras foram mantidas em estufa para germinação de sementes e monitoradas a cada 15 dias por seis meses. Ao todo, 494 indivíduos pertencentes a 19 famílias foram amostrados, distribuídos em 30 gêneros e 40 morfoespécies. A diversidade e a distribuição das espécies no fragmento é homogênea, podendo ser usado como uma fonte de propágulos para restauração florestal.
